Isaac Holman was son of Isaac and Mary Holman. He married Catherine Wilcox on March 20, 1781, in Rowan County, North Carolina. They had eleven children: Daniel, Rachel, Elizabeth, Edith, Isaac, William, Mary, Aaron, Moses, Catherine, and Mahala. After Catherine died in Clark County, Indiana, he traveled back to Rowan County, North Carolina and married Lillis Mitchel on August 24, 1804. They had two children: Matilda and Andrew. 

During the Revolutionary War, he enlisted as a private soldier in Rowan County, North Carolina for eight months under Captains Samuel Reed and Ritchie Grimes. He fought in the battles of both Cowans Ford and Gullford Courthouse in North Carolina. In 1799, he moved to Woodford county, Kentucky. A year later, he moved to Clark County, Indiana, where he obtained a 250 acre land grant on May 18, 1801. He was one of the Clarksville property appraisers and was appointed supervisor in 1802.
